Stacking Camps up to 4 times is quite achievable. 5 or more times on Netural Camps can be a nightmare though depending on the Camp.

Seriously though, in order to achieve it, you only need to be able to judge 3 things:-
First of all, how much HP the Creep needs to drop to before your Attack will secure the Kill
How much Damage it will take from your Lane Creeps
The timing of your Last Hit based on your animation + Attack Speed
Depending on Base Attack Damage and animation, some Heroes are easy to do this with, others can take a lot of practise, i.e. Crystal Maiden, Io, etc.

There will be always something that goes wrong - 2 creeps being evenly attacked simultaneously, mouse slip, lack of damage, highground miss chance on mid, etc.
I think it's a bit overrated to practise getting every lasthit, as it's only really probable when you're getting complete freefarm. For example, when playing in the midlane you usually don't get that much out of it as the challenge is easier if you attack when you are sure to get the lasthit and not immediately when you can get the lasthit. (Let's say you're playing QoP and have 60 damage. It's easier when the creep is at 30 hp, while you could lasthit it at 50 hp) Of course I'm not saying it's useless to practise it but there's more to lasthitting than just freefarming.
I'd consider having a sense for where you can farm a much more important aspect.

You can see this happening a few seconds before you have to land the Last Hit, just throw out an auto-attack or 2 to lower one of the Creeps so you have enough time to last hit both of them.

...then you're doing it wrong.
You should be aiming for the Last Hit as soon as you can. You should never wait for the Creeps HP to drop low enough before starting your Attack animation. You should start the Attack so that it finishes as soon as the Creep's HP is low enough. This minimizes your opponent's chances of Denying your Creeps and gives you more time to set up for the next Last Hit.
If you can't time your Last Hits like this...then you really need to practice...8{(

You have no damage, remember? You can easily hit a creep and last hit it, and then swith on another creep, but with minimal damage you won't able to do enough blows because you also have to watch other creeps(i.e one creep is low-hp and has t be hit soon, but there are other two rapidly dropping in health at the same time).

Of course it isn't, last hitting a creepwith ~20(or so, I don't remember) damage isn't impossible too, but in kobold challenge 10+ lh+denies is already a very solid result.

Not mentioning that creep equilibrium will screw up naturally and with your help, if you don't want creeps to end up under one of the towers, you will have to sacrifice some last hits fro equilibrium eventually most of the time.

Then why you are giving advice if you don't even play Solo Ranked? It's not like any of those thing relate to your ability/disability to play RMM. It's essentially the same as saying "well get on a 5k player level and then go ranked even if you are in sub-1k dumpster" aka "git gud".
Isn't the point of practicing last hitting challanges like that not to ensure that you can hit all of the CS (You need more than just practicing last hits to be able to achieve that in either safe lane or mid), but instead to make last hitting second nature so you never have to actively focus on doing it?
Since, being physically capable of getting all the last hits in mid makes little to no difference if the enemy has the ability to control the lane better than you in mid. And it's a similar case for the safe.
